Hello, I had booked a ticket from Shinjuku to Kawaguchiko station. Is it possible that I can get off at Shimoyoshida station before the bus stop at Kawaguchiko station? If it can't, please suggest me some advice. Thank you.

Sure you can get off at Shimoyoshida. However, please be noted that once you get off, you cannot get back on the bus from Shimoyoshida to Kawaguchiko. You can take a train from Shimoyoshida to Kawaguchiko.
